    Depression

    How it works

    Bupropion is a selective inhibitor of the neuronal re-uptake of catecholamines (noradrenaline and dopamine) with minimal effect on the re-uptake of indolamines (serotonin) and does not inhibit either monoamine oxidase. The mechanism of action of bupropion as an antidepressant is unknown. However, it is presumed that this action is mediated by noradrenergic and/or dopaminergic mechanisms.

    Side Effects

    Agitation, dry mouth, insomnia, headache/migraine, nausea/vomiting, constipation, and tremor, taste disorders, dizziness, concentration disturbances.

    Drug Interactions

    Inhibitors/inducers of the CYP2B6 isoenzyme (e.g., orphenadrine, thiotepa, cyclophosphamide, ticlopidine, and clopidogrel) , paroxetine, sertraline, norfluoxetine, and fluvoxamine as well as nelfinavir and efavirenz , antidepressants (SSRIs, many tricyclics), beta-blockers, antiarrhythmics, and antipsychotics , nortriptyline, imipramine, desipramine, antipsychotics e.g., haloperidol, risperidone, thioridazine , beta-blockers (e.g., metoprolol), antiarrhythmics e.g., propafenone, flecainide , MAO Inhibitors , Levodopa and Amantadine , antipsychotics, other antidepressants, theophylline, systemic steroids, Alcohol.

    Indication

    It is indicated for the treatment of major depressive episodes.

    When not to Use

    It is contraindicated in patients with hypersensitivity to bupropion or any of the excipients , In patients with a current seizure disorder or any history of seizures , In patients with a known central nervous system tumour.

    Zylexx Sr Precautions

    Precaution

    All patients being treated with antidepressants for any indication should be monitored appropriately and observed closely for clinical worsening, suicidality, and unusual changes in behavior, especially during the initial few months of a course of drug therapy, or at times of dose changes, either increases or decreases.
